In Australia, in the past, pasture legumes were rotated mainly with cereals, but increasingly these rotations now involve pasture legumes with a wider range of crops, including legumes. This increasing frequency of the leguminous host in the rotation system may be associated with increased root rots in legumes in the current pasture-crop rotations. The primary aim of this study was to see whether the pathogenicity on pasture legumes of strains of Rhizoctonia solani sourced from lupins and cereals (common crops in rotation with pastures) is associated with increased incidence of root rots in pasture legumes in the disease conducive sandy soils of the Mediterranean regions of southern Australia. The second aim was to determine sources of resistance among newly introduced pasture legumes to R. solani strains originating from rotational crops as this would reduce the impact of disease in the pasture phase. Fifteen pasture legume genotypes were assessed for their resistance/susceptibility to five different zymogram groups (ZG) of the root rot pathogen R. solani under glasshouse conditions. Of the R. solani groups tested, ZG1–5 and ZG1–4 (both known to be pathogenic on cereals and legumes) overall, caused the most severe root disease across the genotypes tested, significantly more than ZG6 (known to be pathogenic on legumes), in turn significantly >ZG4 (known to be pathogenic on legumes) which in turn was >ZG11 (known to be pathogenic on legumes including tropical species). Overall, Ornithopus sativus Brot. cvs Cadiz and Margurita, Trifolium michelianum Savi. cvs Paradana and Frontier and T. purpureum Loisel. cv. Electro showed a significant level of resistance to root rot caused by R. solani ZG11 (root disease scores ≤1.2 on a 1–3 scale where 3 = maximum disease severity) while O. sativus cvs Cadiz and Erica showed a significant level of resistance to root rot caused by R. solani ZG4 (scores ≤1.2). O. compressus L. cvs Charano and Frontier, O. sativus cv. Erica, and T. purpureum cv. Electro showed some useful resistance to root rot caused by R. solani ZG6 (scores ≤1.8). This is the first time that cvs Cadiz, Electro, Frontier, Margurita and Paradana have been recognised for their levels of resistance to root rot caused by R. solani ZG11; and similarly for cvs Cadiz and Erica against ZG4; and for cvs Charano, Erica, and Electro against ZG6. These genotypes with resistance may also serve as useful sources of resistance in pasture legume breeding programs and also could potentially be exploited directly into areas where other rotation crops are affected by these R. solani strains. None of the tested genotypes showed useful resistance to R. solani ZG1–4 (scores ≥2.0) or ZG1–5 (scores ≥2.5). This study demonstrates the relative potential of the various R. solani ZG strains, and particularly ZG1–4, ZG1–5, ZG4 and ZG6 to attack legume pastures and pose a significant threat to non-pasture crop species susceptible to these strains grown in rotation with these pasture legumes. Significantly, the cross-pathogenicity of these strains could result in the continuous build-up of inoculum of these strains that may seriously affect the productivity eventually of legumes in all rotations. In particular, when choosing pasture legumes as rotation crops, caution needs to be exercised so that the cultivars deployed are those with the best resistance to the R. solani ZGs most likely to be prevalent at the location.  相似文献

克隆植物根据其构型可以分为游击型和密集型。游击型克隆植物的间隔子长,分株在水平空间的扩展范围大,可以利用更大空间范围内的资源,其通过克隆生理整合作用发生的非局部反应的能力强。由此可以得出的推论之一是,小生境斑块的环境发生变化,生长于其中的密集型克隆植物的反应可能会更灵敏。这种反应可能会体现在生物量以及配置格局的变化上。以毛乌素地区沙生半灌木群落中两种优势克隆植物羊柴(H edy sarum laeve)和油蒿(A rtem isia ord osica)为研究对象,前者是典型的游击型克隆植物,后者是密集型克隆植物。采取野外调查的方式,观测在不同植被盖度的小生境斑块内二者地上生物量分配格局的变化情况,并结合二者的克隆构型和生活史特征试图探讨产生这种格局的原因。结果表明:羊柴的地上各部分生物量对植被盖度变化的响应不如油蒿敏感。这或者是因为羊柴的游击型克隆构型决定其可以跨越小尺度斑块实现克隆生理整合,可以利用不同小生境斑块的资源导致的。油蒿只能利用小生境斑块内的资源,当小生境斑块的条件改变,其生物量以及配置方式也随之发生相应的变化。在繁殖方式上,羊柴的有性繁殖结构以及有性繁殖投资显著小于油蒿。在资源有限的条件下,对一种繁殖方式的投资常常会削弱另一种繁殖方式。羊柴主要依靠克隆繁殖,这或者符合并支持配置理论的观点。  相似文献

目的:了解泰山沙参属植物资源现状,测定其根中脂肪、蛋白质及多糖的含量,为合理开发利用泰山沙参属植物资源提供依据。方法:采用野外实地调查法进行资源考察;分别用索氏提取法、考马斯亮蓝法和苯酚-硫酸法测定脂肪、蛋白质和多糖的含量。结果:采集的100余份标本,经鉴定为沙参属植物狭叶沙参[Adenophora gmeli-nii(Spreng)Fisch.]、石沙参(Adenophora polyantha Nakai)、杏叶沙参(Adenophora stricta Mig.)及细叶沙参(Adeno-phora paniculata Nannf.)。泰山沙参属植物狭叶沙参、石沙参、杏叶沙参和细叶沙参脂肪含量分别为2.14%~7.34%,4.27%~7.72%1,.54%~2.51%和4.98%。蛋白质含量分别为0.60~2.10 mg/g0,.80~1.89 mg/g,0.83~0.89 mg/g和1.05 mg/g,多糖含量分别为20.58%~63.21%2,7.74%~65.14%,43.14%~48.47%和45.60%。结论:泰山野生沙参属植物资源丰富,品种多、分布广、蕴藏量大,多糖含量较高,具有较大的开发前景。  相似文献

以小麦品种济南177悬浮细胞系来源的原生质体与同品系胚性愈伤组织制备的原生质体混合后作为受体;以经过380μW/cm2紫外线照射1min、2min的新麦草原生质体分别作为供体,用PEG法诱导融合。组合Ⅰ(176+cha9+新麦草UV 1min)获得16个再生克隆。经过形态学、同工酶、染色体和RAPD分析,确定其全部为属间体细胞杂种。其中的5个克隆再生杂种植株。用7对小麦SSR引物对杂种克隆的叶绿体基因组进行了分析;组合Ⅱ(176+cha9+新麦草UV 2min)只获得3个克隆,且逐渐褐化死亡。表明以小麦济南177的两种培养细胞混合作受体的融合体系有利于杂种的获得及再生;紫外线对融合产物的生长发育有明显的剂量效应。  相似文献

Shoot tips of Amembranaceus excised from in vitro grown axillary bud were encapsulated in calcium alginate beads. Subsequently, shoot tips were precultured in liquid MS medium enriched with 075mol·L-1 sucrose for 5d at 25℃ and then desiccated aseptically on dried silica gel for 5h to a water content of 231% (fresh weight basis) prior to immersion in liquid nitrogen (LN) for 1d. After rewarming at a 40℃ water bath for 2-3min and transferred to solid culture medium for shoot tip recovery. About 50% of cryopreserved shoot tips grew into shoots within 2 weeks after plating. Cryopreservation of Astragalus membranaceus (Fisch.) Bge. shoot tips by encapsulation vitrification has also been developed. Excised shoot tips were firstly encapsulated into alginate gel beads and then precultured in liquid MS medium containing 1mg·L-1 6 BA, 005mg·L-1 NAA and 075mol·L-1 sucrose at 25℃ for 3d. After loading for 90min with a mixture of 2mol·L-1 glycerol and 04mol·L-1 sucrose at 25℃, shoot tips were dehydrated with PVS2 for 120min at 0℃ prior to direct immersion in liquid nitrogen for 1d. After rapidly thawing at a 37℃ water bath for 2-3min, shoot tips were washed for 10min with liquid MS medium supplemented with 1mg·L-1 6 BA, 005mg·L-1 NAA and 12mol·L-1 sucrose at 25℃ and then post cultured on solid MS medium supplemented with 2mg·L-1 6 BA, 005mg·L-1 NAA. The regeneration rate of shoot tips amounted to nearly 80%. Both of plantlets regenerated from cryopreserved shoot tips were morphologically uniform, which both showed as that of control plants. Thus, this encapsulation dehydration and encapsulation vitrification technique appears promising as a routine method for the cryopreservation of shoot tips of Amembranaceus.  相似文献

Potentilla parvifolia Fisch . (Rosaceae) is a traditional medicinal plant in P. R. China. In this study, seven flavonoids, ayanin ( 1 ), tricin ( 2 ), quercetin ( 3 ), tiliroside ( 4 ), miquelianin ( 5 ), isoquercitrin ( 6 ), and astragalin ( 7 ), were separated and purified from ethyl acetate extractive fractions from ethanol extracts of P. parvifolia using a combination of sevaral chromatographic methods. The human neuroblastoma SH‐SY5Y cells were differentiated with all trans‐retinoic acid and treated with okadaic acid to induce tau protein phosphorylation and synaptic atrophy, which could establish an Alzheimer's disease cell model. The neuroprotective effects of these flavonoids in cellular were evaluated in vitro by this cell model. Results from the Western blot and morphology analysis suggested that compounds 3 and 4 had the better neuroprotective effects.  相似文献

2015年7月, 沿荒漠植物花棒(Hedysarum scoparium)天然分布带, 在内蒙古、宁夏、甘肃选取7个典型样地, 采集花棒根际0-30 cm土层样品, 研究丛枝菌根(AM)真菌群落结构变化和土壤因子的生态功能。试验基于形态特征对分离的AM真菌孢子进行分类鉴定, 共分离鉴定AM真菌6属42种。群落结构分析表明不同样地的AM真菌群落结构差异显著, 土壤因子对AM真菌群落有重要影响。由东到西随干旱程度加剧, AM真菌种丰度、孢子密度、Shannon-Wiener指数逐渐降低。共有种孢子密度由东向西逐渐降低, 但在群落中所占比例逐渐增加。同一样地不同AM真菌种属、不同样地同一种属的AM真菌孢子密度不同。相关性分析结果显示土壤有机碳、pH值、氨态氮和有效磷对AM真菌影响显著, 土壤湿度对AM真菌影响极显著。结果表明花棒根际AM真菌群落结构和物种多样性具有明显的空间异质性, 并与土壤因子关系密切, 其中土壤湿度对AM真菌生态分布的影响最明显。  相似文献

确定了制备甘草酸分析样品的超声波提取条件, 0.3%稀氨水为溶剂,液固比50:1,提取时间5 h;确定了高效液相色谱法检测甘草酸含量的条件为ODS色谱柱(4.6mm×25 cm, 5 μm),检测波长254 nm,检测温度为室温,流动相CH3OH/3% HOA c(V/V)为75/25,流速1mL/m in,进样量10 μL,平均加样回收率100.20%,相对标准偏差为1.77%。结果表明超声提取-高效液相色谱法是一种准确度高,速度快的测定甘草中甘草酸含量的检测方法。  相似文献

不同程度的沙埋是生长在干旱和半干旱区内陆沙丘的植物经常遭遇的事件,沙埋可以改变植物所处的生物和非生物环境条件。已有研究表明不同程度的沙埋对于植物的影响不同。轻微程度的沙埋可以增加植物高度、促进生物量的积累和新生分株的产生。如果沙埋强度不断增加,对植物的影响由正效应逐渐转变为负效应。即超过一定沙埋阈值后,沙埋会削弱植物的生长,甚至影响植物的存活。干旱和半干旱区内陆沙丘中常常生长着许多克隆植物,克隆整合常常可以缓解克隆植物分株所遭受的局部环境胁迫。根茎型克隆植物羊柴(Hedysarum laeve)是毛乌素沙地的优势半灌木之一,也是当地重要的固沙植物。为了探讨克隆整合的作用是否可以提高沙埋阈值,并有助于羊柴忍受高强度的沙埋,以其为研究对象开展了野外实验。结果表明:轻微程度的沙埋(例如沙埋深度是原始羊柴分株高的10%~20%)可以加速羊柴分株的高生长,提高叶片生物量、茎生物量以及整个地上部分的生物量。高强度的沙埋(例如沙埋深度是原始羊柴分株高的80%~100%)会削弱羊柴分株的存活和生长。在与不遭受沙埋分株相连的情况下,羊柴分株遭受沙埋的阈值高于没有分株相连的,而且在高强度的沙埋下,前者(有分株相连的遭受沙埋的分株)比后者(没有分株相连的遭受沙埋的分株)在株高增量、茎生物量、叶片生物量以及地上分株生物量上都要显著高。这暗示着克隆整合提高了羊柴遭受沙埋的阈值并有助于羊柴分株忍受高强度的沙埋。  相似文献

利用大型环境生长箱研究了两种幼龄沙地优势灌木柠条 (Caraganaintermedia) 和羊柴 (Hedysarummon golicum) 对CO2 浓度倍增和土壤干旱交互作用的响应。CO2 浓度倍增并没有改善两种沙生灌木叶片的水分状况, 而土壤干旱使叶片的相对含水量 (RWC) 显著降低。在土壤水分充足条件下, CO2 浓度倍增促进两种沙生灌木植株生长, 在干旱条件下则主要促进根的生长, 提高根冠比。土壤干旱显著减少了植株生物量, 但相对促进了根的生长, 特别是显著提高了羊柴的根冠比。CO2 倍增使稳定性碳同位素组分 (δ13 C) 降低, 但土壤干旱使之增加。两种沙生灌木叶片与根部的δ13 C值呈极显著线性关系, 羊柴的斜率大于柠条的, 表明前者叶片与根部在光合产物分配上具有较高的生态可塑性, 这和干旱条件下羊柴的根冠比增加相关联。羊柴的“源库”调节特性反映了对土壤水分胁迫具有较高的耐性。  相似文献
